I went to Niagra Falls 16 years ago.
Driving will take 9 hours from NYC.
You will also need to cross into Canada if you want to get the real experience of Niagra Falls. Americans now need to take their passports! It's ridiculous.
I don't know any tour operators up there, since we never used one. Check the internet, there must be at least a dozen.
Of course you will need to stay overnight, at least 2-3 nights.
Niagra Falls has everything right there. Lots of entertainment, use a taxi or walk. You can also take a bus ride to Toronto. I forgot how long it took from Niagra. Maybe an hour+?
Good luck.

Take the Amtrak from Penn Station and get off at Niagara Falls. Take a taxi to your hotel :)
Depending on your Amtrak accomodations, you can get one with a sleeping cabin/room or just the regular coach.

7-8 hours drive (unless the conditions are really bad). And winters are NOT a good idea since it can get really really cold!! Consider yourself warned :)
But if you are up for the adventure... make a point to go (and spend more time) on the Canadian side, the views are far better from their side (since you are looking *at* the falls, and not looking *down* at the falls) (plus night life is much better there). The picture perfect "horseshoe falls" is barely visible from the american side, and thats a must see... especially for a photographer... A good option on the American side (and a must do, if they allow it in the winters) is to go down to the very bottom of the falls... it's an amazing experience....
If you are travelling alone, I would reccommend flying instead of driving, unless you wanna capture some shots on your way (it's semi scenic, offering some really nice landscapes). No need to get a tour operator, the place is small, and you have the freedom to explore on your own...
Depending on who you are travelling with, you dont need more the 2 nights of stay...
Dont worry about the local transportations, you wont really need any, the town is really small, and most of the hotels are 10-15 mins walk from the falls.. (on either side).
Hope this helps.

Our itinerary a few years ago looked like this:
Day 1: Fly from New York/La Guardia to Buffalo, take a bus from their to our hotel on the Canadian side.
Day 2: Spend all day walking around the falls, take a boat tour; entertainment at night.
Day 3: Left hotel in the morning, bus/flight back to La Guardia.
I remember that eating, entertainment, and everything was within walking distance. Except for the bus to/from the airport we didn't use any other transportation.

I'm originally from Buffalo wich is a 20 min. drive from the falls . Trust me, if you drive you risk losing your car and your life. Its not uncommon being stranded in a car overnight in a blizzard that can dump 7 ft. of snow. Pack extra clothes if you fly since its possible to be stuck at the airport a couple of days.
The good news is the falls turns into an amazingly beutifull ice palace in winter and is well worth the trip. Check out Goat Island on the US side. Definitly see the Canadian side. The Canadian side is more internationally friendly and modern. Be VERY cautious w/ the border crossing into Canada AND returning to the USA . Research what the currant boarder crossing policy is. Since 911 legally they can do what ever they want . Good luck . You will definitly get some unbelievable shots . See Niagara Falls link below .

I was there Sept.06.
It is COLD up there in the Winter!
Water is splashing all around, and if you get wet, you get pneumonia, and you die.
Go there no later than Sept. Wait till 11pm when the lights are turned on. check out my gallery for pics.

I did this in one day about 20 years ago when my grandparents came to visit. We flew up (probably to Buffalo), then taxi-ed it to the falls. We did not go over to the Canadian side but we got a pretty nice view, nevertheless. It's doable in a day, but an overnight trip might be a nice getaway from the city.
